Analysis of Dexamethasone in Jamu Pegal Linu Circulating in Ciracas Market Using TLC-Spectrophotodensitometry

Abstract

Jamu is a traditional medicine widely used for health maintenance, health promotion, and disease prevention. One of them is jamu pegal linu, which is efficacious in relieving muscle and bone pain. However, the increasing trend of jamu consumption is accompanied by the risk of adulteration with Chemical Drug Additives (BKO) such as dexamethasone. Long-term use of jamu containing dexamethasone can resulting adverse health effects include stomach ulcers, allergic reactions, hypertension, diabetes, liver and kidney damage, and can even be fatal. This study aims to identify the dexamethasone content in jamu pegal linu circulating in Pasar Ciracas, East Jakarta. The method used was a qualitative test using Thin Layer Chromatography (TLC), with the results determined by spot visualization and the comparison of Retention Factor (Rf) values. The TLC results of 10 jamu pegal linu samples showed that 1 sample (sample 3) formed a spot aligned with the dexamethasone reference standard when viewed under UV 254 nm light. The Rf value of sample 3 (0,538) was identical to the Rf value of the dexamethasone standard. The process continued with sample spot scanning, which showed that only 1 sample (sample 3) had a chromatogram spectrum aligned with the dexamethasone standard, while the other samples did not. Based on the scanning results at a maximum wavelength of 243 nm, the chromatogram spectrum of sample 3 was identical to the dexamethasone standard. This confirms that sample 3 is positive for dexamethasone. Thus, it can be concluded that 1 out of the 10 samples studied was proven to contain dexamethasone.
